Tamsulosin is a selective alpha1-adrenoceptor blocker that primarily targets alpha1A adrenoceptors in the prostate. By blocking these receptors, Tamsulosin relaxes the smooth muscles in the prostate and bladder neck, improving urine flow and relieving symptoms of Benign Prostatic Hyperplasia (BPH).
After oral administration on an empty stomach, approximately 90% of Tamsulosin is absorbed. The time to peak concentration (Tmax) is 4–5 hours when taken without food and 6–7 hours when taken with food. Tamsulosin binds extensively to plasma proteins (94%–99%) and is primarily metabolized in the liver by cytochrome P450 enzymes, with less than 10% excreted unchanged in the urine.
The elimination half-life varies based on the formulation:
- Immediate-release formulations: 5–7 hours
- Modified-release capsules (Prostam): 9–13 hours in healthy individuals, and 14–15 hours in patients with BPH
